JetPRIME transfection reagent
A novel non-liposomal reagent based upon the cutting edge delivery technology by Polyplus.
jetPRIME combines powerful transfection of DNA & siRNA with very low toxicity. It uses LOW amounts of nucleic acid and reagent making it versatile and economical and an ideal reagent for day to day transfections.
jetPRIME, the NEW transfection reagent from Polyplus Transfection is highly efficient yet extremely gentle to cells. Advantages ► LOW cytotoxicity ► Excellent cell viability ► Compatible with serum AND antibiotics ► High DNA transfection efficiency ► Excellent gene silencing ► Economical and simple to use Convenient protocol jetPRIME is an easy-to-use transfection reagent: • Fast and easy to scale up and down (Fig. 1) • Simple: 1 : 2 DNA to jetPRIME ratio (w/v) • No medium changes required, hence time saving • Compatible with serum and antibiotics |

Higher transfection efficiency with small amounts of DNA
jetPRIME is such a powerful in vitro transfection reagent that it only requires a small amount of plasmid DNA (Fig.2 and 3). Upon comparison with other transfection reagents, jetPRIME was the most efficient at lower DNA concentrations.
Fig. 2 Transfection efficiency assessed by flowcytometry analysis in various cell lines following transfection of pCMV-GFP plasmid. Transfection was performed using 250 ng DNA and 0.5 µl jetPRIME per well in 24-well plate following the manufacturer ‘s recommendations. |

Outstanding transfection efficiency with the lowest toxicity
jetPRIME reagent shows maximal transfection efficiency in a variety of commonly used cells, using only 2 µg of DNA per well in 6-well plates. Using small amounts of DNA and reagent is not only economical but reduces the cytotoxic effects of using too much DNA. Using jetPRIME reagent and DNA, transfection efficiencies range between 70-90% on standard cell lines. The figure below shows GFP expression in HEK-293 and SK-OV-3 cells 24 h after transfection with pCMV-GF plasmid using jetPRIME.
Gene silencing with jetPRIME
|
jetPRIME leads to over 90% knockdown of endogenous gene expression in a variety of cell lines. For example, jetPRIME-mediated transfection of HeLa cells with10 nM siRNA duplexes targeting endogenous lamin A/C in HeLa cells reduces lamin A/C gene expression to barely detectable level (Fig. left) Endogenous lamin A/C silencing using jetPRIME. HeLa cells were transfected with 10 nM of 21-mer lamin A/C siRNA. After 48 h, lamin A/C silencing was assessed by immunofluorescence microscopy using an antibody against lamin A/C. jetPRIME is also well suited to DNA/siRNA co-transfection. |
Low Toxicity
Phase contrast microscopy of Hela cells 24 hours after transfection according to the manufacturers recommended protocols. Cells transfected using jetPRIME remain healthy while major cytotoxicity is shown with those using the competitor. |
