Q: Why use an siRNA transfection reagent with 1nM siRNA?

A: Because scientists want to obtain results they can rely on- studies show that using low siRNA concentrations avoids the unwanted toxic and off-target effects observed when transfecting siRNA at higher concentrations (Persengiev S et al 2004 RNA 10:12).

Over 90% gene silencing in a wide variety of cells

For adherent cell lines or primary cells, 1 nanomolar or less siRNA concentration is sufficient to give more than 90% gene silencing, regardless of the supressed gene. for difficult to transfect suspension cell lines, 80% silencing can still be reached using 5nM siRNA.

Endogenous Lamin A/C silencing using INTERFERin

CaSki cells were transfected with 1nM of 21-mer siRNA duplexes matching the lamin A/C sequence using INTERFERin. After 48h, lamin A?C silencing efficiency was determined using immunofluoresence microscopy. 

INTERFERin mediated siRNA transfection inhibits luciferase expression in A549-GL3Luc cells

Cells were transfected in 24 well plates in the presence of serum with decreasing concentrations of Luciferase siRNA (GL3Luc) or control siRNA (mismatch GL2luc) duplexes using INTERFERin. Luciferase expression was measured after 48h.
